Purification and Properties of Cellodextrin Phosphorylase from Clostridium-Thermocellum
Cellodextrin phosphorylase [EC 2.4.1.49] was purified 129-fold, with a yield of 22.9%, to electrophoretic and column chromatographic homogeneity from a cell extract of Clostridium thermocellum ATCC 27405 by a procedure which included streptomycin treatment, ammonium sulfate precipitation, DEAE-Toyopearl 650 M, and Toyopearl HW-55F column chromatography. The molecular weight of the enzyme was estimated to be 200,000 by gel filtration and 105,000 by SDS-PAGE, suggesting that it consisted of two identical subunits. It was suggested by spectrophotometric and chemical analysis that the enzyme contained no pyridoxal 5’-phosphate. The enzyme was inactivated by N-ethylmaleimide and activated by dithiothreitol, indicating that the exposed thiol group(s) was important for the enzymatic activity. The enzyme could synthesize at least cellotriose, cellotetraose, and cellopentaose as detectable cellodextrins, showing that it might possibly be a good tool for the synthesis of cellodextrins.
