Development of technology for dapagliflozin production
Technol. process for a total synthesis of amorphous form of dapagliflozin was optimized to increase the temp. of Li-Br exchange with BuLi from -78 degrees C to -30 degrees C. The process consists of 5 chem. stages (including isolation of peracetylated dapagliflozin) and one chem. step connected with amorphization of pure dapagliflozin. The process was successfully implemented under pilot plant conditions with overall yield of 50%.
