Post-synthetic Modification of 3 ' Terminus of RNA with Propargylamine: A Versatile Scaffold for RNA Labeling through Copper-catalyzed Azide-Alkyne Cycloaddition
We report the chemical alkynylation of oligoribonucleotides with propargylamine. Stepwise treatments of oligoribonucleotides with NaIO4, propargylamine, and NaBH3CN yielded N-propynylmorpholine derivatives at the 3' terminus. This RNA labeling approach is applicable to any natural RNA that possesses a ribonucleoside at the 3' terminus.
