PCR cloning of Pseudomonas resinovorans polyhydroxyalkanoate biosynthesis genes and expression in Escherichia coli
A ca. 5.5-kb region of Pseudomonas resinovorans genome containing the polyhydroxyalkanoate (PHA) biosynthesis locus was sequenced. Three complete open-reading-frames (ORFs), i.e., phaCl(Pr), phaZ(Pr), and phaC2(Pr), were identified. Using this sequence information, phaCl(Pr) was PCR-cloned from P. resinovorans genomic DNA and expressed in E. coli as shown by a Nile Red plate assay and gas chromatography/mass spectrometric analysis.
