Simple estimation of Langmuir Parameter by HPLC experiments

HPLC 칼럼을 사용하여 단일성분의 등온흡착 곡선을 구하는 실험을 수행하였으며, 파라미터를 계산하기 위하여 equilibrium-dispersive model을 (Macsyma®(Macsyma Inc., Arligton, MA, USA.)로 전산모사하여 파라미터들을 계산하였다. 전산모사와의 적합성을 검증하기 위하여 시료로 카페인을 사용해 샘플의 과량주입 실험을 통하여 실험치와 계산치를 비교하였으며, 시료의 농도와 이동상의 유속 변화에 따른 용출 피크들의 변화를 관찰하였다.
A simple method of isotherm determination for HPLC is discussed. The equilibrium-dispersive model describing the behavior of elution peaks in HPLC for single component is solved numerically by using PDE solver Macsyma®(Macsyma Inc., Arligton, MA, USA.) and compared to the experimental data obtained in overloaded isocratic chromatography with caffeine as model species. The effect of sample concentration and flow velocity on the band profkles of elution peaks are described.
