Evaluation of thermo-responsive injectable hydrogel containing oxidized cellulose nanofibre for post surgical adhesion prevention |

Of late thermo-responsive and biodegradable injectable hydrogels are gaining attention for post-operative anti adhesion. In this work we investigated the influence of different concentrations of oxidized cellulose nanofibre (OCNF) on prevention of post surgical abdominal adhesions particularly in a rat cecal abrasion model. An effective injectable adhesion barrier was formulated comprised of methyl cellulose, polyethylene glycol, carboxymethyl cellulose and OCNF, with unique inverse thermal gelling, high stability and anti-adhesive properties. The gelation temperature was optimized by adjusting the concentration of Na+ and other components to ensure transformation of solution into gels at body temperature (37oC), wherein gelation time was found to be approximately 120 seconds. Hydrogel containing 0.2 % OCNF showed minimal adhesion of rat bone marrow-derived mesenchymal stem cells on the surface based on confocal and optical images. MTT assay showed that the hydrogels were all non-cytotoxic, and degradation studies revealed that the higher the OCNF concentrations the lower the chance of degradation. In vivo study confirmed that the hydrogel easily adhered to the damaged peritoneal surfaces, and gradually degraded within 14 days. Hematoxylin and eosin staining ensured no adhesion and adverse immune reaction on obtained tissue sections. These results show that OCNF- containing injectable hydrogels has potential for clinical applications as an effective, safe and versatile material on preventing post-surgical abdominal adhesions. |
