제목 Heparin immobilization onto intraocular lens
초록 In this study, it was investigated the effect of surface bonded heparin onto polymethyl methacrylate (PMMA) intraocular lens on cell adhesion to improve the biocompatibility. The PMMA surface was oxidized with ozone, and then heparin was graft-bonded. To verify the heparin grafting on the surface, the sulfur content was measured by electron spectroscopy for chemical analysis. The amount of heparin determined by toluidine blue method can be controlled with reaction conditions. Heparinized surface revealed the decreased dynamic receding contact angles and static contact angles. The heparin grafted PMMA revealed a lower dynamic receding contact angle than the untreated PMMA. The adhesion of HLEB3 cells onto modified PMMA was investigated in vitro and in vivo. Untreated PMMA, heparin-bonded PMMA, control sample inserted in the anterior chamber of a colored rabbit. A commercially available intraocular lens, 811C PMMA (Pharmacia Co., Sweden) was used as a control sample. The mean number of HLEB3 cells onto untreated PMMA were 2.4×105 cells/㎖, however it was decreased to 1.1×105 cells/㎖ onto heparin bonded ones to yield a significant statistical difference (p〈 0.05). In vivo results, they were not difference between the heparin-bonded PMMA IOL and 811C PMMA (Pharmacia Co., Sweden). This result suggests that surface bonding of heparin onto PMMA intraocular lens may reduce retroprosthetic complications in ophthalmic applications.
