Glutamate transporter inhibitors are blockers that inhibit glutamate transport in synaptosomes. The glutamate is the excitatory neurotransmitter in the central nervous system. It is a major constituent of protein in human body, however, over concentration of glutamate is considered one of the reasons of the Alzheimer’s diseases. Therefore, we found that synthesis of glutamate transporter inhibitors is motivating and especially focused on L-threo-β-benzyloxyaspartic acid (L-TBOA). In our previous reports, a synthetic method for threo-β-amino-α-hydroxy acid was introduced. With this method in hand, we synthesized L-TBOA. In this study, synthetic route from D-serine to L-TBOA will be contained. A series of the functionalization including O-benzylation afforded the L-TBOA efficiently. |
