Self-assembled nanoparticles for targeting of CD44 receptor of tumor cells |

Since hyaluronic acid (HA) receptor such as CD44 and RHAMM is over-expressed at sites of cancer cell attachment, HA has key roles in motility, invasion, and metastasis of cancer cells. HA can be used as a targeting device for metastatic cancer cells, because metastatic cancer cells normally over-express CD44 receptor. This study was to examine fabrication of polymeric micelles using block copolymer composed of HA and poly(ε-caprolactone) (HAPCL) and their targeting potential. Synthesis of HAPCL was confirmed using 1H NMR spectroscopy. Polymeric micelles prepared by dialysis method have less than 100 nm in diameter. Doxorubicin as an anticancer drug was incorporated into the core of the polymeric micelles and targeting potential was examined using HepG2 hepatocellular carcinoma cells. When DOX-incorporated polymeric micelles were treated, red fluorescence was found at cancer cells while blocking of CD44 receptor by HA treatment resulted in reduced fluorescence intensity, indicating that HAPCL polymeric micelles have CD44R-mediated targetability against cancer cells. We suggest HAPCL polymeric micelles were superior anti-cancer drug delivery system. |
