제목 | Dextran-based block copolymer for pH-responsive delievry of doxorubicin |
초록 | Nanoparticles based on stimuli-responsive drug delivery system have been extensively investigated in recent decades [1,2]. Among them, pH-sensitive drug targeting using pH-sensitive polymers against solid tumor has attracted remarkably since solid tumor has acidic environment [3]. Tumor targeting based on pH-responsive polymeric nanoparticles or micelles is known to have superior antitumor activity against various solid tumors. In this study, we synthesized block copolymers composed of poly(ethylene glycol) (PEG) and dextran conjugated with histidine (DexHSPEG) to make pH-responsive nanoparticles. As a model drug, doxorubicin (DOX) was incorporated into the nanoparticles of DexHSPEG and nanoparticles were formed by nanoprecipitation/dialysis method. DexHSPEG nanoparticles encapsulating DOX showed increase of particle sizes and drug release rate in the acidic pH. DOX-incorporated DexHSPEG nanoparticles showed higher anticancer activity at acidic pH. |
