Effects of charge densoty of conjugated polymer on florescence resonance energy transfer-based biosensor signal |

Fluorescence resonance energy transfer (FRET) from cationic poly(fluorene-co-phenylene) derivatives to dye-labeled single-stranded DNA (ssDNA-C*) was studied by changing charge density of the polymer repeating unit (RU). Although the molar absorptivity and quantum yield of the polymers in water are increased with increasing the charge density, FRET-induced dye emission is decreased. It is expected that the local concentration of RUs around C* at the saturation of the electrostatic complexation is decreased as the charge density increases. In addition, photo-induced charge transfer (PCT) quenching which is sensitive to the intermolecular distance between polymer and ssDNA-C* is reduced as polymer charge density is decreased due to the weaker complexation. The effect of ionic strength of the solvent on the FRET and quenching process will be also discussed in detail in terms of polymer charge density and electrostatic complex formation. |
