Restoration of damaged endothelium might be an attractive way to reduce late stent thrombosis and restenosis. Outgrowth endothelial cell (OEC) can differentiate to endothelial cell and then facilitate re-endothelialization. Some groups have developed various OEC capture strategies such as monoclonal antibody (Ab), magnetic nanoparticles, and aptamers immobilization technologies for the restoration of endothelium. We focused on CD146 as a target surface biomarker for OEC capture. In this study, it was demonstrated that anti-CD146 Ab-immobilized stent can capture OEC and restore damaged endothelium. Fabrication of silicon nanofilament was accomplished for a number of antibody immobilization. Silicon nanofilament and anti-CD146 Ab-immobilized surface were evaluated by immunofluorescence, FT-IR, and an OEC capturing assay. These results exhibited that anti-CD146 Ab-immobilized surface captured significantly more OECs than bare surface and anti-CD34 Ab-immobilized surface. |
