Induction of re-endothelialization on growth factor-loaded metal surfaces modified with heparin and hyaluronic acid |
초록 |
Rapid re-endothelialization of damaged vessel lining efficiently prevents restenosis and thrombosis and restores original vascular functions. For this purpose, several growth factors, such as vascular endothelial growth factor (VEGF) and hepatocyte growth factor (HGF), have been reported to promote re-endothelialization. In this study, we designed heparin-grafted surfaces for loading those growth factors using 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) or divinyl sulfone (DVS). First, we grafted heparin onto a dopamine-conjugated hyaluronic acid coated Co-Cr surface to serve as a growth factor reservoir. In the second step, we loaded VEGF or HGF on heparin-conjugated Co-Cr surfaces through their heparin-binding domain. It was demonstrated that DVS technique offered higher amount of growth factor loading than EDC. In vitro assessments also showed enhanced capillary-like structure formation and localized gap junctions when DVS coating was employed. |
