Simple diagnostic system for tuberculosis magnetophoretic immunoassay using surface modified Fe3O4 and Au nanoparticles with tuberculosis antibodies |

Fe3O4 nanoparticles (MNPs) and Au nanoparticles(Au NPs) have been widely used in biomedical fields, in particular, serving as diagnostic agents due to unique magnetic and optical properties. In this study, we introduce a simple diagnostic method of tuberculosis (TB) by magnetophoresis using immunological combination of MNPs and Au NPs via the TB antibody-antigen reaction. The surface of two individual NPs was modified with two-types TB antibodies, respectively. These particles from both routes were added to poly ethylene glycol (PEG) solution (5 w/v %) and sonicated for 10 min for additional stable dispersion and the protection from human pH range. These are immersed in the specially designed TB cell culture bottles. When the secretion of TB antigens happens, two different NPs are linked through immuno-reaction resulting in the union of Au and magnetic NPs which is correlated with the quantity of TB antigen. |
