Receptor-Mediated Gene Delivery by Folate-PEG-Baculovirus In Vitro and In Vivo |

Gene therapy has potentials to treat devastating inherited diseases for which there is little hope of finding a conventional cure. Recombinant baculovirus vectors engineered to contain mammalian cell-active promoter elements have been applied to the delivery of genes into mammalian cells. In a previous study, PEG-baculovirus (P-Bac) was prepared to regulate non-specific transduction of baculovirus. However, P-Bac has limitation of targeting to specific tissue. As a targeting ligand, folate has been reported to have a great potential for targeting to the folate receptor (FR). Because the FR expression is frequently amplified in variety of human cancers, it has been suggested to have the potential utility of FR as a cellular marker for cancer-targeted drug and gene delivery. In this study, folate-P-Bac(F-P-Bac) was synthesized and characterized as a receptor-mediated gene carrier. Also, the transduction efficiency in vitro and the profile of tumor progression in vivo were evaluated. |
