제목 Synthetic polymers for non-viral gene delivery system to human turbinate mesenchymal stem cells (hTMSCs)
초록 Gene delivery using non-viral vectors has received a great deal of attention because of ease in preparation, stability, and safety. The aim of this study is to evaluate transfection efficiency hTMSCs using DNA/polyethyleneimine (PEI) complexes. DNA/PEI complexes with N/P charge ratios in the range 1–16 were formed by adding appropriate amounts of PEI to the DNA. The formation of complexes was characterized by gel electrophoresis, measurement of particle size and zeta potential. The viability of hTMSCs was conducted using the MTT assay. After gene transfection, green fluorescence was monitored by microscope and FACscan. DNA was completely condensed with PEI above N/P 1-2. As the portion of PEI was increased, the particle size of complexes decreased, the surface charge and cytotoxicity to hTMSCs were increased. The transfection efficiency was 47% at N/P 8 in 24 hours. Our study demonstrates that PEI may be applied as an effective gene carrier to hTMSCs.
