In tissue engineering and regenerative medicine, adult stem cells and hydrogels are widely used for cell sources and scaffolds, respectively. In this work, we synthesized a methoxy polyethylene glycol-polycaprolactone block copolymer (PCL), and then made a PCL solution. A PCL solution was sol-state at room temperature and gel-state at body temperature. hTMSCs had mesenchymal stem cell markers and proliferated at high rate. Before in vivo experiments, we observed osteogenic differentiation of hTMSCs in vitro by Alizarin Red S, von Kossa and alkaline phosphatase staining. To confirm osteogenic differentiation of hTMSCs in vivo, we mixed hTMSCs and a PCL solution containing osteogenic factors, and then prepared solution implanted to dorsum of nude mouse by subcutaneous injection. We monitored osteoblastic phenotypes and gene expression through histological methods and qRT-PCR, respectively. In conclusion, hTMSCs could serve as adult stem cell sources and tissue engineering applications. |
