Gene therapy is the use of DNA as a drug to cure disease by delivering normal or therapeutic DNA into abnormal cells. Microportaion, virus, liposomes, and non-viral carriers were able to use as gene delivery methods into cells. Polyethyleneimine (PEI) is most common non-viral gene carriers containing high number of amine group in main and side chain. It is affordable and easy to prepare, however, the cytotoxicity of PEI is higher following increase of the amounts. Therefore, we prepared biocompatible polyester gene delivery carriers, PCL-NH2, and compared the cytotoxicity and gene delivery efficiency. We mixed PEI and PCL-NH2 solution with DNA solution respectively, characterized the complexes of PEI/DNA and PCL-NH2/DNA. Although transfection efficiency of PEI is higher than PCL-NH2, the results of cytotoxicity tests revealed that PCL-NH2 is biocompatible and less toxic than PEI. To conclude, PCL-NH2 are confirmed to have potentials as a gene delivery system. |
