제목 Effect of chemical structure and Molecular Weight of PolyethylenimineGene Carrier on the Reverse Transfection-mediated Transgene Expression
초록 Gene delivery by non-specific adsorption of non-viral vectors to protein-coated surfaces can reduce the amount of DNA required, and also increase transgene expression and the number of cells expressing the transgene. Nucleic acids can be complexed with polymer to form DNA nanoparticles which are then immobilized on the surface coating extracellular matrix protein (ECM), termed as reverse transfection. We investigate the effect of molecular weight (Mw) and 3-dimensional structure of positive-charged PEI on transfection efficiency via surface-mediated delivery by changing Mw and structures of PEI. Different types of PEI (branched PEI Mw 25kDa, linear PEI Mw 1.8kDa, linear PEI Mw 10kDa, and linear PEI Mw 25kDa) were examined for enhanced transfection to neighboring cells. Our initial results indicate that branched type displays higher gene transfer and that higher Mw produces better transfection to neighboring cells.
