Effective implantation of outgrowth endothelial cells by extracellular matrix immobilization for promoting re-endothelialization of coronary stent |
초록 |
Reconstruction of collapsed endothelium is prominent for the prevention of in-stent restenosis and late thrombosis. To incorporate the blood vessel and promote angiogenesis, we modified the cobalt chromium (CoCr) stent with polydopamine (pDA) and fibronectin (FN), and constructed cell-derived extracellular matrix (ECM) via our decellularization method. To In vitro cell study, ourgrowth endothelial cells (OECs) were cultured on the ECM immobilized CoCr surfaces. The result showed that the ECM was successfully immobilized on the pDA/FN coated surface, and OECs are well attached on the ECM-immobilized CoCr surfaces compared to bare CoCr. In in vivo study, OEC implantation using ECM stent achieved higher re-endothelialization rates and lower neointima formation rates than BMS. These suggest that the OEC implantation using ECM stent would be an effective therapeutic treatment for accelerating re-endothelialization and suppressing complications. |
