Effect of BHA.PLGA scaffold on the neural differentiation of rat BMSCs |

Bone marrow stromal cellsn(BMSCs) can be differentiated into, chondrocytes, neuronal cells under the controlling of culture condition. We fabricated the BHA(Butylated Hydorxyanisole) loaded poly(L-lactide- co-glycolide) (PLGA) scaffolds (BHA/PLGA) for the purpose of neurogenesis of BMSCs. BHA/PLGA scaffold were manufactured as the concentrations of 200, 400, and 600μM by solvent casting/salt leaching method. We isolated BMSCs from female Fischer rat. Celluar viability and were assayed by MTT test and Morphology of celluar adhesion was confirmed by SEM.The effect of BHA/PLGA scaffold on the neural differentiation of rat BMSCs were assessed RT-PCR and staining. We confirmed that effect of BHA/PLGA scaffold on the neural differentiation of rat BMSCs. According to results, 200, 400μM BHA/PLGA scaffold have higher expression of neural markers. In conclusion, BHA/PLGA scaffold, on which neural differentiation of rat BMSCs was possible. This research was supported by SCRC(SC3100). |
