RNA has been used for producing nano-sized structures but it has not usually synthesized with macroscopic scale. In this study, our group introduced a RNA film by rolling circle transcription without additional supporting materials. The RNA film had free-standing property and its structural and functional properties could be modified by controlling design of RNA sequence. To further research the usage of RNA film, we examined the potential for drug delivery system. We loaded intercalating chemical drug to RNA film and we observed that it could induce controlled release of drug by enzymatic reaction. Furthermore, to test the application for delivering siRNA, we rationally designed RNA film to contain siRNA region. We checked the generation of siRNA by Dicer reaction and it could be used for siRNA therapies. |
