Bioabsorbable nanofibers containing blood coagulation agents for use as hemostatic agents |

Hemostatic agents have been used in surgical operation for many years to control bleeding. In this study, we fabricated poly(vinylpyrrolidone) nanofibers containing calcium chloride and thrombin to use as a novel non-woven hemostatic agent. PVP nanofibrous sheets were fabricated by electrospinning of 10 wt% PVP aqueous ethanol solution containing thrombin and calcium chloride. Electrospun PVP nanofibrous mat structures revealed randomly aligned fibers with average diameter of 580 nm. Surface characteristic of PVP nanofiber were confirmed to be soluble and hydrophilic surface determined by water contact angle measurement. PVP nanofibers showed a relatively low cytotoxicity and the cellular survival rate was more than 90%. In vitro whole blood coagulation experiment showed expeditious blood coagulation within 3 minutes after contact with fabricated nanofibrous sheet. These results suggest that electrospun PVP nanofibers will be a promising hemostatic agent to reduce massive blood loss. |
