The effect of fibrin/PLGA scaffold on the inflammatory reaction. |

PLGA(lactide-co-glycolic acid) (PLGA) has been widely applied to tissue engineering as a good biocompatible material. However, their acidic degradation products can decrease the pH in the surrounding tissue, which result in local inflammatory reaction. The cells neighboring the PLGA implant might have an inflammatory response. We could be reduced by impregnating fibrin into the PLGA. In vitro, cell viability and proliferation analyzed by MTT and WST assay. The release profiles of TNF-α were assayed using ELISA. TNF-α and IL-6 mRNA expression was analyzed using RT-PCR to evaluate the inflammatory response of cells on the fibrin/PLGA scaffolds. In vivo, inflammatory cells and fibrous wall thickness were evaluated by H&E and ED-1 staining. In the results, inflammatory marker showed lower in fibrin/PLGA scaffold than PLGA scaffold. This study shows that fibrin of PLGA could reduce inflammatory reaction of PLGA. This research was supported by WCU (R31-20029) and SCRC (SC4100). |
