제목 | Comparativeevaluation of in vitro transcribed Firefly and Gaussian luciferase mRNAexpression in cells |
초록 | Luciferaseis often used as a reporter protein to evaluate target protein expression. Ithas various advantages such as high sensitive and easy of detection, short half-lifeto investigate the protein expression profile. Typically, Firefly and Gaussianluciferase are used to evaluate the cytosolic luc expression in cells and/orsecreted luc expression in medium. Firefly and Gaussian have different ORFlengths of 2kb for firefly and 500nt for Gaussian. Depending on use ofdifferent luciferase, quantitative analysis of mRNA based protein expressioncan be investigated. In this study, both of Firefly and Gaussian luc mRNA wereencapsulated in the lipid nanoparticles and treated to cells to monitor thetime dependent expression of reporter proteins. Our study showed bothquantitative and comparable assay method to evaluate the luciferase expressionin cells. |
