Magnetic particles are one of the most popular materials and have been increasingly applied to data storage, magnetic resonance contrast-enhancing media, immobilization of proteins, and biotechnology. However, bare magnetic particles may not be very useful in practical applications, since they easily form a large aggregation, and alter magnetic properties when they are directly exposed to the biological system. Here, we will introduce the novel method for surface treatment by layering magnetic particles with alternate positive and negative charge of polyelectrolytes, and present their usage for simple bio-application. These amine or carboxylic surface-modified magnetic particles by adsorption of polyelectrolyte are well-dispersed and highly stable in water. Onto these magnetic particles, probe oligonucleotides are chemically conjugated and then complementary target oligonucleotides labeled with Cy5 are hybridized. Fluorescence images are obtained by confocal microscopy. |
