The homeostasis of labile zinc is intimately associated with a variety of pathophysiological states in the human body. Although numerous molecular zinc sensors have been created, most of them are not suitable for imaging zinc-rich specimens due to the propensity to bind zinc tightly. We have developed a new fluorescence sensor (HBO-ACR) exhibiting fluorescence turn-on responses with low affinity to zinc ions (zinc dissociation constant (Kd) = 60 μM). HBO-ACR shows selective responses to Zn over Cd and Hg ions and other competing biological metal ions. The fluorescence zinc responses are not affected by protonation, providing large turn-on ratios over wide pH ranges. Finally, bioimaging capability of the zinc probe has been successfully demonstrated by employing PANC-1, human pancreas cells. |
