Self-assembled Micelle-type Cationic Nanoparticles Based on Polyethlenimine- Poly(lactic-co-glycolic acid) Conjugate for Gene Delivery |

During last decades, a lot of synthetic gene carriers have been proposed for non-viral gene delivery. Among the synthetic polymers, polyethylenimine(PEI) is one of the most widely used non-viral gene carriers. In this study, an amphiphilic PEI-abased coploymer conjugate was synthesized. Poly(lactic-co-glycolic acid) (PLGA), an NIH-approved biodegradable polymer, was employed as a hydrophobic partner of the conjugate. The chemical structure of the conjugate was determined by using H-NMR analysis. The copolymer formed micelle-like aggregates with critical association concentration (cac) in 2.55×mg/ml. The cationic nano-aggregate could interact with plasmid DNA via ionic force, resulting in the formation of polyelectrolyte complexes with a size ca. 200 nm. The self-assembled nano-aggregate having a micelle-like core-shell structure could be used as a functional vehicle for simultaneous delivery of therapeutic genes and water insoluble drugs. |
