제목 Conjugation of Vitamin and Cationic polymer and Its Application for Non-vial Gene Delivery
초록 In order to improve the transfection efficiency of non-viral vector and to reduce the cytotoxicity of cationic polymer, the cationic polymer and vitamin conjugates were synthesized by carbodiimido method. They were confirmed by UV-spectrophotometer and TNBS method, and characterized by gel electrophoresis, particle size and zeta potential to estimate a possibility as a gene carrier. In the case of PRA/DNA complex, a completed complexation was observed above 3 of N/P ratio, while cationic polymer/DNA complex showed above 2. The zeta potential of the conjugates decreased with increasing vitamin concentration in PRA conjugates. The transfection efficiency of PRA in HeLa cell was up to 27 times higher than that of cationic polymer system. The cytotoxicity of PRA was also measured by MTT assay. PRA conjugates showed lower cytotoxicity compared with the cationic polymer system. Therefore, the PRA conjugate may be good candidate for non-viral gene carrier.
