Promoting of re-endothelialization by recruiting angiogenic cells on anti-CD146 antibody immobilized stent with silicone nanofilament |
초록 |
Bare metal stent (BMS) is widely utilized for treating coronary artery diseases but BMS causes restenosis by vascular smooth muscle cell hyperplasia. Therefore, a new concept of stent has been developed to recruit circulating endothelial progenitor cells (EPCs) by immobilized antibody (Ab). Previously developed CD34 Ab coated stent seemed to have an effect on circulating EPCs capturing. However, it is thought that the CD34 Ab coated stent also causes restenosis because the capturing capability of CD34 Ab has still controversy. Here, we targeted CD146, a specific transmembrane protein of late EPCs, as a biomarker for capturing late EPCs. In addition, we introduced silicone nanofilaments (SiNf) to increase surface area. By virtue of increased Ab number, SiNf-CD146 Ab surface showed higher OEC-capturing ability than bare and flat surfaces in in vitro. In vivo study using porcine restenosis model showed the modified stent enhanced the re-endothelialization and decreased restenosis rate. |
