Efficient Gene Delivery Systems Using Inverted Quasi-spherical (iQS) Culture Substrates for Cell-gene Therapy |

In the field of cell and gene therapy, the genetically modified therapeutic cells have been studying for efficient medical approaches using viral or non-viral gene delivery vehicles. Using these approaches, the efficiency of gene delivery and expression levels are crucial for increasing therapeutic effects. Herein, the inverted quasi-spherical (iQS) system, which were fabricated with the hydrophilic dot patterns on the TiO2 coated superhydrophobic substrates, provided the steep curvature of hanging droplets during three-dimensional (3D) cell cultures. This unique microenvironment enhanced the cell-cell and cell-gene interactions. In this study, the human neural stem cells (hNSCs) were showed the enhanced transduction efficiency of adeno-associated viral vectors (AAV) and increasing of protein secretion levels of therapeutic cytokine using this iQS culture systems. |
