Smart Bead-based Platform of Immunoassay Using a Stimuli-responsive Biopolymer and a Removable Polymeric Template |

We present a smart bead-based platform of immunoassay using an elastin-like polypeptide (ELP) conjugated microbeads and a removable polymeric template. To make a defined bead microarray, micropatterns of a removable polymeric template were transferred onto the spun-cast adhesive thin layer by microcontact printing. UV-curable adhesive was applied to enforce the adhesion of the microbeads at the substrate and its adhesion performance was controlled by UV irradiation time. The microarray of polystyrene bead conjugated with ELP was used for the smart immunoassay of prostate specific antigen (PSA), a cancer marker. By controlling the phase transition of ELP molecules, PSA molecules were selectively captured and separated from the mixture of biomolecules. This method can be successfully applied to a variety of substrates including metal, glass, polymers and ceramic to fabricate single or multiple microbead arrays. |
