Here we demonstrate that myocardial protection of infracted heart using dual growth factor delivery with self-assembling peptide nanofiber hydrogel (SAP), RADAII-16. Myoardial infarction (MI) was produced in ~ 150g SD rat. After left coronary artery ligation, growth factor with SAPNS was injected into infracted border zone (n=6 in each group: Sham, MI, MI + SAP, MI + SAP/FGF-2, MI + SAP/PDGF-BB, MI + SAP/FGF-2/PDGF-BB, and FGF-2/PDGF-BB). The hearts were harvested 4 weeks after for functional and histological analysis. Infarct area was 22.3 ± 3.6 % in MI + SAP/FGF-2, 17.3 ± 0.7 % in MI + SAP/PDGF-BB, and 8.5 ± 2.2 % in MI + SAP/FGF-2/PDGF-BB. Ejection fraction was 30.7 ± 0.6 %, 34.1 ± 0.8 %, and 43.7 ± 1.4 % in each group (compared to 18.37 ± 2.9 % in MI, 51.0 ±1.8 in sham). In this study, dual growth factor delivery with SAP is effective to myocardial protection and leads to improvement in cardiac function. |
