제목 Hyaluronic acid-modified liposomal nanoplatform for NIR-responsive chemo-photothermal treatment of rheumatoid arthritis
초록 The combination of photothermal therapy (PTT) and chemotherapy regarded an effective treatment for diseases such as cancer and inflammation. Herein, we developed a nanoplatform named HMM-Lipo, a NIR-responsive hyaluronic acid (HA)-modified liposome containing methotrexate (MTX) and melanin. Due to HA modification, HMM-Lipo targeted specifically activated CD44 receptor in rheumatoid arthritis (RA). HMM-Lipo was spherical with an average diameter of 131.9±9.7 nm. Upon NIR irradiation, heat was generated and MTX release from HMM-Lipo was accelerated. Cell blocking experiments showed that HMM-Lipo was selectively internalized through CD44 receptor-mediated endocytosis. When HMM-Lipo was injected into the tail vein of collagen-induced arthritis (CIA) mice and exposed to a NIR laser (0.5 W/cm2, 3 min), swelling and redness of the inflamed area were alleviated. Thus, HMM-Lipo may provide high potential as an RA therapeutic agent for NIR-responsive chemo-photothermal therapy.
