A series of blended natural polymeric scaffolds composed of β-chitosan and gelatin was prepared for bone tissue regeneration. Scaffold was crosslinked and hydrolyzed by genipin and 1N NaOH. BMP-2 immobilized scaffold was prepared by reacting EDC-activated hydroxyl group of scaffold and amine group of BMP-2. It was observation that the series of scaffold (non-treated, hydrolyzed, and BMP-2 immobilized scaffolds) could cell viability and osteogenic differentiation of DPSCs. DPSCs were seeded in the porous scaffold, and evaluated by the cell viability, ALP activity, alizarin red assay. The series of porous scaffolds did not affect cell growth because of low cytotoxicity. We observed that BMP-2 immobilized scaffold was a significant induction of ALP activity, alizarin red when differentiated osteogenic from DPSCs. These results indicated that BMP-2-immobilized scaffold would be valuable for bone tissue regeneration. |
