An in vitro assessment of BMP-2 and GDF-5 loaded Photo-crosslinkable hydrogel conjugated with zirconium surface for enhanced osseointergration |

BMP-2 and GDF-5 loaded photo-crosslinkable hyaluronic acid hydrogel was immobilized on a zirconium (Zr-HA gel) surface. The characterizations of non-treated Zr and Zr-HA gel with or without BMP-2 and GDF-5 were assessed by contact angle and XPS. MG63 cells were seeded and cultured on the series of Zr surfaces, and then osteogenic activities as alizarin red, alkaline phosphatase activity, and RT-PCR were compared at 7, and 14 days. The surface of Zr immobilized with photo-crosslinkable hydrogel showed more hydrophilic surface than that of non-treated Zr. After the culture of 7 days, osteogenic activities on Zr-HA gel with or without BMP-2 and GDF-5 were found to be higher than those on non-treated Zr. After 14 days, osteogenic activities from BMP-2 and GDF-5 loaded Zr-HA gel were found to be significantly higher than non-treated Zr and Zr-HA gel. The BMP-2 and GDF-5 loaded Zr-HA gel surface may be taken into consideration in designing Zr implants to improve the status metal implant. |
