In vivo Chondrogenesis of BMSC in Injectable Hyaluronic acid/F127 Hydrogel with Dexamethasone-loaded PLGA Microsphere |
초록 |
Hyaluronic acid (HA) is a major component of the ECM in the connective tissues. It has unique physicochemical properties and distinctive biological functions in cartilage. and binds specifically to the proteins in the cartilage ECM. Pluronic F127 is a thermosensitive synthetic polymer. Both materials are considered biocompatible. Growth factor-loaded microspheres have been utilized as a delivery system, in which they would promote differentiation of specific cells. In this study, Dex-loaded PLGA microsphere is encapsulated in F127/HA composite hydrogel with bone marrow-derived stem cell (BMSC). We hypothesize that this system may be favorable in inducing chondrogenesis of BMSC in an injectable mode. The cell-loaded hydrogels are implanted into nude mouse subcutaneously. After 2 and 4 weeks, the constructs are evaluated using histological staining and RT-PCR for cartilage-specific gene expression. |
