Polymeric and liposomal layer-by-layer coating for efficient delivery of nitric oxide: Application to coronary stent |
초록 |
Restenotic and thrombotic drawbacks of stent use is the reason why many types of stent have been produced up to date. In this study, we focused on trying to develop a coating method for efficient NO delivery in which utilizes liposome containing a NO donor and biocompatible polymers, which can achieve a long-term sustained release of NO by comparing three coating techniques including a) coating on a metal, b) coating on a polymer, and c) layer-by-layer (LbL) coating. Among these, only the LbL coating accomplished a sustained release of NO for over 5 days when compared to the other two techniques. In addition, biological experiments revealed that the least fibrinogens adsorbed on the LbL as compared to controls, and endothelial cell proliferation also increased for 7 days. In vivo experiment showed that NO-LbL stents have positively significant differences in the neointimal area and the percent area of stenosis when compared to the bare metal stent (BMS) group. |
