Dual Stimuli-responsive poly(N-isopropylacrylamide)-block-poly(L-dihydroxyphenylalanine) Copolymer Micelles for Theranostic Delivery |

We prepared a series of diblock poly(N-isopropylacrylamide)60-block-poly(L-dihydroxyphenylalanine)n (p(NIPAM)75-b-p(L-DOPA)n) (n=35,50,75,100) polymer bio-conjugates via combination of reversible addition fragmentation chain transfer and ring opening polymerization of α-amino acid N-carboxyanhydrides. All the diblock polymer forms spherical micelles in aqueous solution and thus provides enough space for accommodate the potent anticancer drug Doxorubicin (Dox). Micelle size and morphology are studied using Transition electron microscopy. Polymer micelles exhibited excellent drug loading capacity and drug loading efficiency, also in vitro drug release profile demonstrated that the maximum release of Dox was happened in pH 5.0 at 37 oC intracellular mimic environment. The controlled Dox release was investigated in different temperature and pH conditions. The pH and temperature induced drug release from the micelle is confirmed by the cellular uptake and cell cytotoxicity study. |
