제목 Preparation of technetium-labeled Galactosylated Chitosan Nanoimaging for Liver Disease Diagnosis
초록 The need to develop target-specific MRI contrast agents to aid in disease characterization remains highly essensial. Macromolecular contrast agents due to their larger size enable longer imaging times and possess higher relaxation times. These agents have been used to enhance imaging of tumor site via active targeting. Parenchymal liver cells, hepatocytes, are useful target cells for imaging and mammalian hepatocytes are the only cells that possess large numbers of high affinity cell-surface receptors that can bind asialoglycoproteins. Therefore, 99mTc-labeled galactosylated chitosan nanoparticles were developed and characterized as liver-targeted nanoimaging carrier. Chitosan was coupled with the galactose group for liver targeting using EDC/NHS method and then labeled with 99mTc for nanoimaging. Labeling efficiency of99mTc to galactosylated chitosan (GC) having various molecular weights was examined using thin layer chromatography (TLC). Particle sizes of 99mTc-labeled GC were determined by light scattering measurements. To examine the labeling stability of the 99mTc-labeled GC nanoparticle, radioisotope-labeled nanoparticle was incubated and the radioactivity of the nanoparticle was measured. Liver specificity and biodistribution of 99mTc-labeled GC nanoparticles were investigated. GC specifically binds asialoglycoprotein receptors of hepatocytes through receptor-mediated mechanism. GC showed very high labeling efficiency of Tc stably for extended time, as determined by TLC. The enhanced liver targeted localization of 99mTc-labeled GC nanoparticle was observed.
