Glycol Chitosan-Based Nanocomplexes as a Sustained Delivery System of Exendin-4. |

Exendin-4, a long-acting GLP-1 agonist, has considered to be more effective than native GLP-1. However, the clinical effectiveness of exendin-4 has been limited by its short biological half-life. Therefore, the long-acting formulation of exendin-4 still remains an interesting challenge. Owing to strong binding of lithocholic acid to ß-cyclodextrin(ß-CD), it was expected that supramolecular systems based on ß-CD could further extend the drug’s duration of action. To comprehend our concept, we have synthesized glycol chitosan bearing ß-CD(ß-CDGC) conjugate and investigated their complexation behavior with LAM1-Ex4. The physicochemical characteristics of nanocomplexes were investigated. Further, db/db mice were administered a single subcutaneous injection of LAM1-Ex4/ß-CDGC nanocomplexes and blood glucose levels were monitored using a glucometer and tail-tip blood samples. The results indicate that nanocomplexes had a potential as a long-acting formulation of exendin-4. |
