Gene delivery with a non-viral carrier to adult stem cells |

In this study, we evaluated the transfection efficiency of polyethyleneimine (PEI) as a gene carrier in two kinds of adult stem cells—rat bone marrow mesenchymal stem cells (rBMSCs) and rat muscle-derived stem cells (rMDSCs). The results of agarose gel electrophoresis identified that DNA is fully bound to PEI at the N/P charge ratio 4. The surface charge of the DNA/PEI nanoparticles was positive at the N/P charge ratio greater than 4. According to the results of cytotoxicity assay, the viability of rBMSCs and rMDSCs was inversely proportional to the N/P charge ratio of DNA/PEI nanoparticles. The fluorescence imaging of cells showed EGFP expression clearly in both kinds of stem cells transfected with DNA/PEI nanoparticles. More specifically, rBMSCs showed high transfection efficiency (1-17%) whereas rMDSCs showed relatively lower efficiency (1-5%). Our results demonstrate that rBMSCs were more effectively transfected with DNA/PEI nanoparticles than were rMDSCs. |
