제목 Local and Sustained Delivery of Dual Growth Factor (bFGF/NGF) for Urethral Sphincter Muscle and Nerve Regeneration
초록 We, previously reported the heparin-Pluronic (HP) nanogels-embedded gelatin-poly(ethylene glycol)-tyramine (GPT) hydrogel prepared using horseradish peroxidase (HRP)-catalyzed reactions of aqueous GPT solutions containing HP nanogels for tissue regeneration. In this study, dual growth factor (bFGF/NGF) delivery using hybrid hydrogel system has been performed and in vitro evaluations have been studied. FGF loaded HP nanogels were prepared by direct dissolution method. After bFGF encapsulation, the average size of the HP nanogels was increased from 90 to 150 nm. We prepared GPT which was containing bFGF loaded HP nanogels and NGF using simple mixing. Growth factors were sustainedly released from the gel matrix over 1 month and the released growth factors amounts could be controlled by changing growth factor loading amounts. The obtained results support that our dual growth factor-carrying hybrid hydrogel system have great potential in regenerating urethral sphincter muscles and nerves.
