제목 | Chitosan sponge as a scaffold for skin tissue engineering and a dermal substitute |
초록 | It was the aim of this study to evaluate chitosan sponge as a scaffold for skin tissue engineering and a dermal substitute in burn wound. The used chitosan sponge made from the chitosan (CS) or the acetylated chitosan (ACS) was coated with 0.1 % type I collagen or not. Deep second degree burn (3X6 cm) was created on the back of pig (n=11) and the full-thickness skin defects were made after 7 days. Integra, Terudermis, CS, ACS, CS coated with type I collagen, and ACS coated with type I collagen were immediately grafted in the wound bed. On day 14 after surgery, 0.025 inch-thick auto-skin was grafted on them. Grafts were evaluated grossly and histologically on day 7, day 14, day 21 after surgery. In all cases, the wounds were closed by the reconstruction of neodermis and successful take of auto-skin graft. Angiogenesis in the upper region of artificial dermis was observed within 14 days after overlaying of artificial dermis. After auto-skin graft, the rete ridges structure was well constructed in all case. The fragments of CS and ACS with or without Type I Collagen coating were present only in the upper regions of neodermis. In summary, this study suggests that the chitosan sponge will be a scaffold for skin tissue engineering and a dermal substitute to replace the collagen based dermal substitute.
