제목 Preparation and Characterization of Porous Scaffold by Novel Paraffin Particles Leaching Method
초록 A novel paraffin particles leaching method for the fabrication of porous, and biodegradable PLGA scaffold has been proposed for the application to tissue engineering. Previous study, emulsion freeze drying method could impregnate with water soluble factors in scaffold but it had trouble in controlling porosity and pore size.
To control more precise porosity and pore size, solvent casting salt leaching method has been widely used. This method creates desired pore size ranges by incorporating sieved salt particles of a desired size and varying the loading ratio of particles to the polymer. However, significant loss of water soluble factors (e.g., drugs, protein, growth factor) incorporated into the scaffold during fabrication would occur during the leaching out process. So we made an attempted novel paraffin particles leaching method. PLGA scaffolds using novel paraffin particles leaching method with porosity range of 45 to 98 %, median pore size range of 20 to 94 ㎛ with larger pore diameters greater than 200 ㎛, and specific pore area in the range of 40 to 278 (㎡/g) were manufacture by varying processing parameters as ratio of PLGA/paraffin and paraffin size. These results suggest that mass loss of paraffin particles leaching processing can confirm residual paraffin.
