Recent advances in tumor targeted drug delivery using nanoparticle (NP) as carrier have demonstrated the potential to cure disease with minimal side effects compared to conventional therapy. However, existing platforms suffer from low intracellular delivery and escaping efficiency from endo/lysosome. To design an improved drug delivery and endo/lysosome escaping, we adopted pH sensitive polymeric NP with photosensitizer and polysaccharides loading anticancer drug. We made polysaccharide backbone with pH sensitive moiety and then made nanoparticles with photosensitizer. This system involves a self-assembly and being sensitive with low pH in tumor microenvironments to promote drug delivery and endo/lysosome escaping. Therefore, this strategy shows selective cellular uptake, endosomal escaping capability and efficient drug targeting. |
