제목 Cross-linked Hyaluronic Acid with 3D-printed Porous Polylactide Scaffolds for 3D Cell Culture
초록 Three dimensional (3D) cell culture systems, which mimic a cell’s in situ environment in tissues, have great potential for overcoming the inherited limitation of two dimensional cultures. Here we introduce a cell-loaded hyaluronic acid (HA) hydrogel network cross-linked via catechol-ferric ion complexation within mechanically stable, open porous polylactide (PLA) scaffolds for 3D tissue culture. 3D PLA scaffolds were fabricated by a fused-deposition-modeling 3D printing technique. HA was chemically grafted with catechol groups and injected into the PLA scaffolds that were pre-treated with ferric chloride to generate a cross-linking network of HA. The loading efficiency of HeLa cancer cells was 89.2 % without significant cytotoxicity. We expect that our approach to generate a 3D hybridized structure will provide a promising materials platform for 3D culture of various tissues.
