제목 | Effects of gamma irradiation on BCNU release pattern from BCNU loaded PLGA wafer |
초록 | Carmustine (BCNU, 1,3-bis(2-chloroethyl)-1-nitrosourea) is an effective chemotherapeutic agent to treat brain tumor. Poly(D,L-lactide-co -glycolide) (PLGA) offers site-specific drug administration without the need for device retrieval. The major objective of this study was to evaluate the effect of different gamma irradiation doses on the release kinetics from BCNU loaded PLGA wafers. PLGA used in this study was a 50:50 copolymer with 8,000 molecular weight. BCNU loaded PLGA wafers were prepared with or without exipient by direct compress method. The wafer and PLGA were subjected to 0, 1.5, 3, 4.5, 6 and 7.5 Mrad doses of gamma irradiation. In vitro release of BCNU from the wafers was assayed by high performance liquid chromatography (HPLC). The effect of gamma irradiation on PLGA molecular weight was evaluated by gel permeation chromatography (GPC). Differential Scanning Calorimetry (DSC) was performed on untreated and gamma irradiated PLGA. The molecular weight and Tg of PLGA decreased with increasing the irradiation doses. But there was no effect on the release pattern of BCNU from BCNU loaded PLGA wafer as soon as irradiation sterilization. Study on the aging effect after irradiation is in progress.
