제목 | Modofication of RGD to MPEG-PCL and its effects on osteogenic differentiation of MSCs |
초록 | Mesenchymal stem cell (MSC) transplantation has showed up as a promising therapy for regenerative medicine. However, it is limited by low grafted cell retention and survival within the damaged tissues. We investigated the effects of Arg-Gly-Asp (RGD) modified methoxy poly(ethylene glycol)-poly(caprolactone) (MPEG-PCL). The RGD Cell adhesion to RGD is mostly mediated by two integrin receptors –α5β1and α2β1. Over a 3 day observation period, cell proliferation was increased when cells were cultured into MPEG-PCL-RGD. Moreover, expressions of α5β1and α2β1 were upregulated in MPEG-PCL-RGD. These results demonstrate that RGD immobilized MPEG-PCL performs synergistically with MSCs to enhance osteogenic differentiation. These findings suggest potential use for RGD-modified MPEG-PCL as a scaffold in the field of tissue engineering and bone regeneration.
