제목 | Preparation and characterization of self-assembled paclitaxel-conjugated MPEG-PCL nanoparticles |
초록 | In this study, PTX-conjugated methoxy poly(ethylene glycol)-b-poly(ε-caprolactone) (PTX-MPEG-PCL) was utilized to improve the water-solubility of PTX. MPEG-PCL was prepared by ring-opening polymerization of poly(ε-caprolactone) (PCL) with methoxy polyethylene glycol (MPEG, 750 g/mol) as an initiator. The hydroxyl terminal group of MPEG-PCL was substituted to a carboxyl terminal by succinic anhydride (SA), after which PTX was conjugated to MPEG-PCL by esterification via condensation reaction, confirmed by 1H NMR. PTX-MPEG-PCL was self-assembled above 2.8 × 10-5 mg/ml analyzed by fluorescence spectroscopy. The self-assembled PTX-MPEG-PCL showed a spherical morphology with a size distribution ranging from 40 to 460nm (DLS and TEM).
